目前主要人類的共同祖先,在七萬年到十萬年前約二千人,從南非先後移居到中
東,繼西向歐洲,東向亞洲,最慢三到一萬年前,越過白令海峽而到美洲或散布在南太平洋群島上。人類在不同地方困難的求生繁殖,至今已逾七十二億,龐大的人口讓我們更了解地球不同資源的不足。歷史上不同動物被獵殺或馴化,不同植物被採收,或種植而為最重要的食物。而菇類的食用、藥用或毒性也被廣泛的利用。
公元前五世紀的希臘詩人,歐里庇得斯(Euripides, 480 – 406 BC)的妻子及三個孩子
吃毒蕈去世。奧古斯都大帝,他指定的繼承人、姪兒 Marcellus 及其孫子 Gaius, Lucius
Caesar 先後被他的妻子莉薇雅(Livia Drusilla, 55BC-29AD)所毒殺,而使莉薇雅和她前夫的兒子 Tiberius 為皇帝。克勞狄(Claudius)皇帝在公元 54 年被其妻阿格里匹娜(Agrippina) 用毒蕈毒殺,而使阿格里匹娜與前夫所生的兒子尼祿(Nero Claudius Caesar)為皇帝。她們所用的毒藥應是瓢蕈(Amanita phalloids, 中毒症狀似飲酒過量,服用後六到二十四小
時引起嘔吐等嚴重胃腸道症狀,經過一段靜止期,繼之肝腎肺衰竭,低血糖、頭痛、神智不清而死亡)。至今吃有毒的蕈,仍時有報導。但是,印度最古老的吠陀經、梨俱吠(Rig
Veda)所述的 Soma 就是會引起幻覺的蠅蕈(A. muscaria)。而從蠅蕈,德國藥理學家 Schmieaeberg 在 1869 年分離到蠅蕈素(muscarine),所生產的神經系統作用稱為類蠅蕈素作用(muscarinic effects),這知識在神經藥理學上佔重要的作用。無毒的菇類是重要的食物,而其中所含不同成份的生理生化作用,使得菇類也是重要的健康食品,甚至新藥開發的對象。 台灣的牛樟芝,過去很多學者,包括這次參與研討會的專家都有貢獻及興趣。本人與同事周正仁、黃瑞齡、盧美光、汪貴珍等研究員、李一宏醫師、農業試驗所張東柱研究員及台大許瑞祥教授篩選出抗 B 型肝炎病毒的牛樟芝菌種,對其 DNA 加以定序,及做一些生物活性研究,而後續研究也在不同學術機構,學校進行中。野生牛樟芝是昂貴的,大多數人無法享用。細胞、組織培養或生物基因工程可以幫我們不依賴空間及時間,在嚴謹條件下,生產品質一致的產品,才可能被使用者信賴,進行臨床療效評估成為藥品。
目前有關牛樟芝培養方法、化學成分及生物活性的報導很多,並進入對發炎體影響的層次。本次研討會能在中研院由生化所的系統生物學專家陳水田博士召開研討會,獲不同單位協助,且有大陸學者參與,是牛樟芝研究向前的重要一步,讓我們共同努力。 敬祝健康

牛樟芝研究與發展之近況

Recent research and development of Antrodia cinnamomea 吳永昌 Yang-Chang Wu
 
Medicinal mushrooms have attracted much attention recently owing to their potent therapeutic activity, especially as chemopreventive and immunomodulatory agents. Antrodia cinnamomea is a treasured Taiwanese mushroom that has been used by aboriginal tribes for centuries to treat food intoxication and to enhance liver functions. It was included in Asian folk medicine in the last few decades with remarkable results in treating inflammatory disorders, cancers, hypertension and hepatitis. This myriad of therapeutic activities encouraged several research groups to subject A. cinnamomea to intensive biological and phytochemical investigation, leading to the isolation of different classes of pharmacologically active secondary metabolites. The in vitro and in vivo biological results of the mushroom extracts and its active components revealed their potent cytotoxic, anti-inflammatory and hepatoprotective activities. The aim of this study is to review recent reports on the biological activities of A. cinnamomea extracts and its active components; quality control protocols; synthetic methodologies for the preparation of active components; developed culture techniques; phylogenetic analysis and gene cloning. This study also tackles major challenges facing future expansion of A. cinnamomea production. 吳永昌

本實驗依據衛生福利部健康食品安全性評估規範,進行國芝生物科技股份有限公司 100%牛樟芝子實體粉末產品之基因毒性試驗與 90 天亞慢性餵食毒性試
驗。基因毒性試驗分為,(1)沙門氏菌(Salmonella typhimurium) TA98、TA100、 TA102、TA1535 及 TA1537 菌株回復突變之 Ames 試驗,試驗結果顯示,100 % 牛樟芝子實體粉末對沙門氏菌回復突變之 Ames 試驗並不具有致變異性,對細
菌基因突變測試結果為陰性反應(non genetic mutation in Ames test)。(2)測試對嚙齒類動物體內(Micronuclei assay in vivo)週邊血液微核發生之比例,藉以評估
直接或間接引發紅血球染色體或有絲分裂之基因變異造成傷害程度。試驗結果顯示,100%牛樟芝子實體粉末各劑量組對小鼠週邊血液之網狀紅血球和網狀紅血球中微核數目均與陰性對照組無顯著性差異,試驗結果為陰性。(3)細胞染色
體變異試驗(Chromosomal aberration test)結果顯示 100%牛樟芝子實體粉末對體
外哺乳類細胞株 CHO-K1 之染色體不具致變異作用。動物毒理試驗包括:(4)大鼠口服急毒性試驗,大鼠(SD 品系)經口單次投予 2 g/kg body weight,投予後觀
察 14 天。試驗結果顯示,處理組大鼠皆無毒性症狀或死亡,100%牛樟芝子實
體粉末對大鼠之口服急毒性 LD50 值大於 2 g/kg body weight。(5) 食用安全劑量值對大鼠 90 天餵食毒性試驗,以 250 mg/kg body weight 每日經口餵食大鼠,連續 90 天。結果顯示各組大鼠皆無毒性症狀,各組之體重、飼料消耗量、尿液學、血液與生化及全身臟器之組織病理檢查等均無明顯與試驗物質有關之組織病理變化。綜合以上試驗結果顯示,100%牛樟芝子實體粉末食用安全劑量值 250 mg/kg bw,經連續餵食大鼠 90 天後,對大鼠無不良反應。

實驗動物毒性病理判讀與量化評估
The qualitative and quantitative evaluations of toxicopathology in  the laboratory animal studies

摘要: 
實驗動物毒理學之研究數據,可提供食品、藥毒物及化學產品對人體、動物及環境生態安全性相關資訊,並探討對生物體傷害之作用機制,適時作為臨床應用及管理參考依據。國際上已建立各種短期及長期之動物試驗規範,經由檢測實驗動物之各項生理值及病理學的判讀評估,直接或間接的證實藥毒物使用之安全性。但毒理試驗之操作或毒性病理判讀方式不當,會影響試驗結果之正確性,不可忽視。毒性病理學為一結合毒理學及病理學之科學,除需具有獸醫病理學對各種疾病及病變描述等專業知識外,尚需熟悉不同實驗動物及各種毒理試驗規範。同時,毒性病理判讀需達到完整、精確及一致性,且能分辨自發性與毒性病變之不同,綜合試驗物質在整體試驗中造成病變與否,為判讀臨床前動物安全性試驗重要證據之一。肉眼及組織病理觀察與記錄,為動物毒理試驗之最後階段。促成精確的毒性病理判讀,需有完整的樣品採集、正確的組織臟器固定保存、良好的組織修片、切片製作品質及染色。一般例行性切片染色常使用蘇木素及伊紅染色,蘇木紫可染出細胞核及核仁等鹼性物質使呈藍色,伊紅則對酸性物質具親合性,可將細胞內蛋白質染成紅色。利用此二種染劑,可區分出正常細胞結構、形態、大小及組織排列。毒性病理判讀之半定量分析首先需使用共通病理診斷專有名詞描述,依組織病變不同加以區分等級,並列表進行組間積分比較。對於難以區分細胞性或細胞來源之病變,可用特殊化學染色法,標定具特殊性細胞,如結締組織或纖維化,利用Sirius red染色法可染出紅色區,藉此可偵測細胞內存在部位及量的變化。另外,細胞膜抗原容易受到福馬林溶液固定之破壞,常以新鮮標本冷凍切片處理,但因細胞形態固定不佳,病變不易判讀。經福馬林液固定後石臘標本染色,雖因抗原性遭破壞,但仍可用組織免疫化學染色法,利用加熱回復抗原特性,將特殊抗原於封臘之組織切片重新再表現。即利用初級抗體覆蓋組織切片一段反應時間後,以次級抗體處理,再利用呈色物質標識出抗原位置。如常用:增殖性細胞核表現抗原 (PCNA)輔助判定增生細胞。對於一般認為病理判讀常有主觀意識,可藉由影像處理系統定量分析並提供較客觀數據加以修正。因此,毒性病理之正確判讀,除可善用組織病變不同之半定量描述分析外,亦可應用影像處理系統之數位病理切片,可客觀加以輔助,兩者結合後可使病理報告內容更加準確,達到動物
毒理試驗最終目的,訂定「無毒害作用劑量值(NOAEL)」之參考依據。

中國大陸牛樟芝研究的概况 林志彬

截止 2014 年 3 月中國知網(CNKI)中國期刊全文數據庫收錄農業、醫藥
期刊的牛樟芝(包括牛樟芝)研究論文 140 篇。包括牛樟芝命名和中醫性味、歸經和功效的研究,牛樟芝的段木栽培技术和液體發酵培养技術的研究,化學成分和有效成分的提取研究,牛樟芝的藥理研究,包括其發酵液不同提取物在体外对人慢性粒细胞白血病细胞株 K562 细胞、人前列腺癌 LNCaP 细胞的抑制作用、促進巨噬细胞釋放 NO 的作用及對叠氮鈉(NaN3)損傷的 PC12 神經细胞的修復作用。牛樟芝對化學性肝损傷的保護作用、體内抗腫瘤作用、免疫增强作用以及大鼠 30 天和 90 天安全性評價的研究。
 
 
Overview on the study of Antrodia camphorata in mainland China

As of March 2014, total 140 articles of Antrodia camphorate(Zhang Zhi) from medical and agriculture journals listed by CNKI data-base. Nomenclative of Zhang Zhi, and the Xingwei, Guijing and efficacy of Zhangzhi by traditional Chinise medicine(TCM), Wood-cultured technology and liquid fermentation technology  chemical composition and active ingredients were investigated.  Pharmacological effects demonstrated that different extracts of Zhangzhi fermentation broth had  antitumor activity on human chronic myelogenous leukemia cells of K562 and human prostate cancer cell line LNCaP in vitro. Zhangzhi extracts also promoted NO release from macrophage and repaired nerve cell PC12 injuried by sodium azide (NaN3). The protective effect of Zhangzhi on chemical liver injury, the immune-potentiating effect and the anti-tumor effect were observed in vivo. Safety evaluation of 30 days and 90 days was studied in rats.
